Isolation to agar

You surface sterilise infected root pieces and incubate them on a suitable agar. After a few days you check the plates for potential pathogens.

Ah ha, fungal colonies are present. You examine these under the compound microscope, using appropriate magnifications. Hyaline fungal structures are visible.
